Overview
Cagrilintide (AM833) is a long-acting acylated analog of human amylin, engineered with fatty acid conjugation to extend its plasma half-life to approximately one week — enabling once-weekly research dosing in preclinical and clinical studies. Amylin is a 37-amino acid peptide co-secreted with insulin from pancreatic beta cells that acts as a satiety hormone through calcitonin receptor (CTR) and amylin receptor (AMY) complexes in the hypothalamus, brainstem, and area postrema.
In clinical research, cagrilintide has been studied both as a monotherapy and in combination with the GLP-1 receptor agonist semaglutide (as the combination "CagriSema"), where the two mechanisms demonstrate additive effects on body weight reduction and glycemic control. Amylin receptor agonism slows gastric emptying, suppresses postprandial glucagon secretion, and reduces food intake through central satiety signaling — pathways that complement and are partially distinct from GLP-1 receptor signaling.
