Overview
Ipamorelin is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH₂) and selective growth hormone secretagogue that acts as an agonist at the ghrelin/growth hormone secretagogue receptor (GHS-R1a). It was developed as a third-generation GHRP following GHRP-2 and GHRP-6, designed specifically to stimulate GH release with minimal off-target endocrine activity. Unlike earlier GHRPs, ipamorelin does not significantly elevate cortisol, prolactin, ACTH, or aldosterone, making it a highly selective research tool for studying the GH axis in isolation.
In preclinical research, ipamorelin produces dose-dependent, pulsatile GH release comparable in amplitude to GHRP-6, while preserving the normal somatostatin feedback loop — preventing GH accumulation to supraphysiological levels. This selectivity profile has made ipamorelin one of the most widely studied GHRPs for research into GH secretion dynamics, body composition changes, bone density, and metabolic regulation.
