Overview
Kisspeptin-10 (KP-10) is the C-terminal 10-amino acid fragment of the 54-amino acid kisspeptin (metastin), the endogenous ligand for the KISS1R (GPR54) receptor. The KISS1 gene product and its receptor are critical upstream regulators of the hypothalamic-pituitary-gonadal (HPG) axis — KP-10 is the biologically active core fragment that retains full receptor binding and signaling activity comparable to longer kisspeptin isoforms.
Kisspeptin neurons in the arcuate nucleus and anteroventral periventricular nucleus project directly to GnRH neurons and act as the master switch for GnRH pulse generation, controlling reproductive activation at puberty, pulsatile LH/FSH secretion, and the preovulatory LH surge. Research applications include reproductive neuroendocrinology, hypogonadotropic hypogonadism models, and studies of the interaction between metabolism and reproduction (kisspeptin neurons express leptin and insulin receptors, linking energy status to fertility).
Mechanism of Action
- High-affinity agonism at KISS1R (GPR54) on GnRH neurons in the hypothalamus, triggering GnRH pulse release
- Downstream pituitary LH and FSH secretion, and downstream gonadal steroidogenesis and gametogenesis
- Integration of metabolic signals (leptin, insulin) with reproductive axis activation
- Regulation of puberty onset timing in research models via kisspeptin neuron maturation
- Potential anti-tumor metastasis activity via KISS1R — original discovery was as a metastasis suppressor gene
