Overview
MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid peptide encoded within the 12S ribosomal RNA region of the mitochondrial genome, making it one of a class of small mitochondria-derived peptides (MDPs) discovered in the past decade. It was first characterized at the USC Leonard Davis School of Gerontology in 2015. MOTS-c is constitutively expressed and secreted by mitochondria in multiple tissues and can translocate to the nucleus under metabolic stress, where it modulates gene expression related to cellular metabolism and stress response.
In preclinical research, MOTS-c has demonstrated exercise-mimetic properties — administration to sedentary mice produced metabolic responses similar to those induced by physical exercise, including improved insulin sensitivity, increased fatty acid oxidation, and resistance to diet-induced obesity. These findings have positioned MOTS-c as a significant subject of research in metabolic disease, aging, and exercise physiology.
